Association between Immunoglobulin and Macrophages in Primary Methylcholanthrene-induced Sarcomas1

چکیده
Ten primary 3-methylcholanthrene-induced murine sarco mas ranging in size from 0.37 to 3.22 ml were selected for studies of tumor-associated immunoglobulin. The purpose of these particular studies was to determine if a significant pro portion of the immunoglobulin was bound to receptors for the Fc portion of Immunoglobulin G molecules on tumor-associated host lymphoreticular cells. The distribution of tumor-associated immunoglobulin was compared to the distribution of macro phages within the tumors using indirect immunofluorescence with specific antisera. Significant amounts of AGi, AG2a,AG2b, and AG3, lesser amounts of AA, and very little AM were ob served. The distribution of each class of immunoglobulin within the tumor was essentially the same, and distribution paralleled the distribution of the tumor-associated macrophages detected with specific antimacrophage serum. Double-labeling studies using fluorescein and rhodamine isothiocyanate-conjugated reagents confirmed that the majority of the immunoglobulin was bound to the same cells that were reactive with the antimacrophage serum. Furthermore, most of the tumor-asso ciated ¡mmunoglobulin, regardless of class, was eluted by washing tissue sections for 5 hr at 37°, a procedure which effectively removes immunoglobulin bound to cellular Fc recep tors. B-lymphocytes or plasma cells were rare in the tumors. Those observations provide strong suggestive evidence that most tumor-associated immunoglobulin in primary murine sar comas is bound to receptors for the Fc portion of Immunoglob ulin G molecules on macrophages, possibly in the form of antigen-antibody complexes.

متن کاملAssociation between Anti-IgA Antibody and Development of Adverse Reactions in Immunodeficient Patients Under Immunoglobulin Therapy
Background: Mild to severe post-transfusion adverse reactions have been reported in a proportion of immunodeficient patients receiving intravenous immunoglobulin (IVIg). Anti-IgA antibody has been proposed to be associated with development of such reactions. Objective: To assess the association between anti-IgA levels of different isotypes and adverse reactions induced in immunodeficient patien...
متن کاملPossible Association of CD68 Positive Macrophages with Some other Prognostic Factors (ki67, ER, PR, Her2 neu) in Primary Breast Cancer and Axillary Lymph Node Metastasis
Background: Breast cancer is formed of a neoplastic component (epithelial) and a non-neoplastic component (stroma). Stromal- stromal and tumor- stromal interactions have been shown in the regulation of cancer cell growth, metastatic capacity and outcome of treatment. Tumor-associated macrophages (TAMs) are a component of tumor stroma reactionsand are considered as an important ...
متن کاملEstablishment and characterization of high- and low-metastatic clones derived from a methylcholanthrene-induced rat fibrosarcoma.
High- (Cl-33H) and low- (Cl-35L) metastatic clones were established from a methylcholanthrene-induced rat fibrosarcoma (FMQ-100). The modal chromosome numbers of the two clones were different. These clones grew in in vitro culture, showing similar growth rate and saturation density. However, in in vivo experiments, Cl-33H exhibited a higher tumor growth rate, tumorigenicity, spontaneous metasta...
متن کاملSeroprevalence of Cytomegalovirus Antibodies by Electrochemiluminescence Method in Young Women Referred to the Clinical Laboratory, Sanandaj, Iran
Background and aims: Maternal primary and recurrent infection of cytomegalovirus (CMV) may be transmitted to the fetus during pregnancy and may have complications such as death or growth, along with the development retardation of the fetus and infant. The aim of this study was to determine the prevalence of immunoglobulin G (IgG) and immunoglobulin M (IgM) antibodies against CMV in you...
